Description

MONJUVI (tafasitamab)

Overview

MONJUVI (tafasitamab) is a CD19-directed cytolytic monoclonal antibody used in the treatment of certain B-cell malignancies. It is designed to target the CD19 protein expressed on the surface of B cells, helping the immune system identify and eliminate malignant cells. MONJUVI is administered as an intravenous infusion under the supervision of a healthcare professional.

Indication

MONJUVI, in combination with lenalidomide, is indicated for the treatment of adult patients with relapsed or refractory diffuse large B-cell lymphoma (DLBCL), including DLBCL arising from low-grade lymphoma, who are not eligible for autologous stem cell transplantation (ASCT). This indication received accelerated approval based on overall response rates observed in clinical studies.

Mechanism of Action

Tafasitamab binds to the CD19 antigen on B cells and promotes the destruction of malignant cells through immune-mediated mechanisms. Its Fc-modified structure enhances immune cell engagement, contributing to increased antibody-dependent cellular cytotoxicity and phagocytosis.

Clinical Evidence

The approval of MONJUVI was supported by the Phase 2 L-MIND study, which demonstrated meaningful and durable responses in patients with relapsed or refractory DLBCL who were not candidates for stem cell transplantation. Clinical data showed encouraging overall response rates and durable remissions in this difficult-to-treat patient population.

Administration

MONJUVI is administered by intravenous infusion according to a defined treatment schedule in combination with lenalidomide. Premedication may be required to reduce the risk of infusion-related reactions. Healthcare providers should monitor patients during and after administration.

Safety Information

Common adverse reactions associated with MONJUVI include neutropenia, respiratory tract infections, fatigue, anemia, diarrhea, thrombocytopenia, cough, pyrexia, peripheral edema, and decreased appetite. Serious risks include infusion-related reactions, myelosuppression, and infections. Patients should be monitored regularly throughout treatment.

Important Considerations

Treatment decisions should be individualized based on patient characteristics, prior therapies, and overall clinical condition. Healthcare professionals should refer to the full prescribing information for complete details regarding dosing, warnings, precautions, and monitoring requirements.
